| Principle | ThisELISAkitusesCompetitive-ELISAasthemethod.Themicrotiterplateprovidedinthiskithasbeenpre-coatedwithADT.Duringthereaction,ADTinthesampleorstandardcompeteswithafixedamountofADTonthesolidphasesupporterforsitesontheBiotinylatedDetectionAbspecifictoADT.Excessconjugateandunboundsampleorstandardarewashedfromtheplate,andAvidinconjugatedtoHorserADIshPeroxidase(HRP)areaddedtoeachmicroplatewellandincubated.ThenaTMBsubstratesolutionisaddedtoeachwell.Theenzyme-substratereactionisterminatedbyaddingStopSolutionandthecolorchangeismeasuredspectrophotometricallyatawavelengthof450nm±2nm.TheconcentrationofADTTinthesamplesisthendeterminedbycomparingtheODofthesamplestothestandardcurve. |
